You start the game as usual, you pick "Adventure". When you're in the lobby, you go to the tutorial door. Then, play all the levels until you get to the final tutorial. Go to the shop, kill the shopkeeper and bomb down the shop floor until you find a hidden 8-bit Spelunker sprite.

Meat Boy	Inside a dark green pod near the end of the Worm.
Purple	Found in a coffin in one of the four main areas.
The Cyclops	Bought from the Black Market for $10,000, can be stolen, or found in a coffin after seen at the Black Market.
The Eskimo	In a coffin in a Yeti Kingdom level.
The Golden Monk	In a coffin the City of Gold.
The Jungle Warrior	Defeat Olmec and get to the exit.
The Ninja	In a coffin in Olmec's Chamber.
The Robot	In a capsule in the Mothership.
The Round Boy	In a coffin in a Tiki Village level.
The Round Girl	In a coffin in a Spider's Lair level.
The Viking	In a coffin in Flooded Cavern or Restless Undead levels.
Van Helsing	In a coffin on the top of the Haunted Castle level.
Yang	Defeat King Yama and get to the exit.
Yellow	In a coffin in one of the four main areas.

Achievement	How to unlock
Addicted	Play Adventure Mode 1000 times.
Big Money	Obtain 500,000 gold.
Casanova	Rescue 10 or more damsels in one game.
City of Gold	Reach the City of Gold.
Eternal Life	Obtain the Ankh.
Good Teamwork	Complete the game with at least two players alive. No shortcuts.
Her Favorite	Win the kapala from Kali.
Ice Creamed	Reach the Temple.
Ironman	Complete the game without using shortcuts.
Jungle Jammed	Reach the Ice Caves.
Low Scorer	Complete the game without collecting any treasure. No shortcuts.
Made It	Complete the game.
Mines Shafted	Reach the Jungle.
Public Enemy	Kill 12 or more shopkeepers in one game.
Seen a Lot	Complete 50% of the journal.
Seen It All	Complete 100% of the journal.
So It Begins	Beat the Tutorial.
Speedlunky	Complete the game in under 8 minutes. No shortcuts.
The Entire Gang	Rescue all 8 hidden characters.
To Hell and Back	Complete the game the hard way.
