You can unlock additional heroes by collecting Hero Stones. These are rewarded after Boss battles and Elite missions. Some can also be gained from Crusades and Trial missions. They are appear at random. The easiest way to unlock a fourth hero is by completing the Elite campaigns repeatedly and watching video advertisements so you can open two chests at once.
