For the mission "The Guns of Leone" you take from JD O'Toole in Portland island. After you've got to the blip on the radar for the sniper rifle that's on top of a roof. Just jump down from the roof and shoot them. (if you do you want to try just sniping them instead. you can hold L while in snipe mode to make it easier to snipe.)
